Target Information
IRS-1 (Insulin Receptor Substrate 1) is a major substrate of the insulin receptor, phosphorylated in response to insulin, IGF-1, and IL-4 stimulation. It is phosphorylated on serine, threonine, and tyrosine residues across various tissues. Casein kinase II mediates part of the insulin-stimulated phosphorylation, with Thr502 and Ser99 identified as key phosphorylation sites. IRS-1 contains three putative 14-3-3 binding sites (Ser270, Ser374, Ser641), with Ser270 located within the phosphotyrosine binding domain responsible for interaction with the insulin receptor.
