Product Specific Information

Description:
The M1/69 monoclonal antibody reacts with the mouse CD24 molecule, also known as Heat Stable Antigen (HSA). This 35–50 kDa molecule is anchored to the plasma membrane via phosphatidylinositol and is expressed by erythrocytes, thymocytes, peripheral lymphocytes, and myeloid lineage cells. CD24 is variably glycosylated, leading to heterogeneity in molecular mass across cell lineages. Antibodies to CD24 show subtle staining differences among lymphocyte populations. CD24 expression helps resolve stages of B lymphopoiesis in mouse bone marrow and may interact with P-selectin (CD62P).

Applications Tested:
Tested by flow cytometric analysis of mouse splenocytes. Recommended usage ≤ 0.125 µg/test. A test is defined as the amount (µg) of antibody staining a cell sample in 100 µL volume. Cell number may range from 10⁵–10⁸ cells/test. Optimal titration is recommended for best performance.

Light Sensitivity:
This tandem dye is sensitive to photo-induced oxidation. Protect vial and stained samples from light.

Fixation:
Samples can be stored in IC Fixation Buffer (Product #00-8222-49) or 1-step Fix/Lyse Solution (Product #00-5333-54) for up to 3 days at 4°C in the dark. Clone-specific performance should be determined empirically.

Excitation / Emission / Laser:
488–561 nm excitation; 775 nm emission; compatible with Blue, Green, and Yellow-Green lasers.

Filtration:
0.2 µm post-manufacturing filtered.

Target Information

CD24 is a two-chain glycosylphosphatidylinositol (GPI)-anchored glycoprotein expressed at multiple stages of B-cell development, from pro-B cells in bone marrow to mature surface Ig-positive B cells. Plasma cell expression is low or absent. CD24 is expressed on most B-lineage acute lymphoblastic leukemias, B-cell CCLs, and B-cell non-Hodgkin’s lymphomas. It may regulate B-cell proliferation, maturation, and autoimmunity control.
