This P4A10 monoclonal antibody reacts with canine CD25, the 55-kDa interleukin-2 receptor alpha (IL-2Rα) chain. In dogs, CD25 is expressed by early T and B lineage progenitors as well as activated mature T and B lymphocytes. CD25 associates with IL-2Rβ (CD122) and IL-2Rγ (CD132) chains to form the high-affinity IL-2 receptor. On its own, CD25 binds IL-2 with low affinity. Along with CD4 and Foxp3, CD25 serves as a major marker for regulatory T cells.

The P4A10 antibody recognizes the same epitope as the anti-human CD25 monoclonal antibody ACT-1 but with greater affinity.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and tested by flow cytometric analysis of normal canine peripheral blood cells. Recommended use at 5 µL (0.125 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Light Sensitivity: This tandem dye is sensitive to photo-induced oxidation. Protect from light.
Fixation: Samples can be stored in IC Fixation Buffer (Product #00-8222-49) or 1-step Fix/Lyse Solution (Product #00-5333-57) for up to 3 days at 4°C in the dark with minimal impact on brightness and FRET efficiency. Clone-specific performance should be determined empirically.

Target Information

CD25 (IL2 receptor alpha chain/IL2RA) is a cytokine receptor involved in T and B lymphocyte proliferation. The IL2 receptor complex (IL2RA, IL2RB, IL2RG) forms the high-affinity IL-2 receptor. Homodimeric IL2RA forms a low-affinity receptor, while IL2RB homodimers form a medium-affinity receptor. IL2 expression in mature thymocytes is monoallelic, regulating precise gene expression. IL2, primarily produced by mature T cells, functions as a growth and differentiation factor and regulates cell death. It stimulates B cell proliferation, enhances NK cell activity, and inhibits granulocyte-macrophage colony formation. Mutations in IL2RA are associated with interleukin-2 receptor alpha deficiency.
