Product Specific Information

The M1/70 monoclonal antibody reacts with mouse CD11b, a 165–170 kDa integrin alphaM. CD11b associates with CD18 to form the alphaMbeta2 integrin (Mac-1) and binds to CD54 (ICAM-1), C3bi, and fibrinogen. It is expressed by macrophages, NK cells, granulocytes, activated lymphocytes, and mouse B-1 cells.
M1/70 also cross-reacts with human CD11b and can detect this antigen on human peripheral blood monocytes, granulocytes, and a subset of NK cells.

Applications Reported: Flow cytometric analysis.
Applications Tested: Tested by flow cytometric analysis of mouse bone marrow cells at ≤0.125 µg/test.
A test is defined as the amount of antibody that stains a cell sample in a final volume of 100 µL. The optimal antibody concentration should be titrated empirically.

Fluorophore Characteristics:
PerCP-eFluor 710 emits at 710 nm and is excited with a 488 nm blue laser. Recommended filter: 710/50 bandpass (695/40 acceptable).
Light-sensitive—protect from light.

Fixation:
Samples can be stored in IC Fixation Buffer (Product #00-8222-49) or 1-step Fix/Lyse Solution (Product #00-5333-57) for up to 3 days in the dark at 4°C with minimal impact on brightness and FRET efficiency.

Target Information

CD11b (integrin alpha-M, ITGAM) is a 165 kDa adhesion molecule that forms a complex with CD18 (integrin beta-2). The CD11b/CD18 heterodimer, known as Mac-1 or CR3, functions as a receptor for ICAM-1, ICAM-2, ICAM-4, CD14, CD50, CD23, heparin, iC3b, fibrinogen, and Factor X.
It is expressed on monocytes/macrophages, granulocytes, activated lymphocytes, NK cells, dendritic cells, and microglia. CD11b is a marker for microglia and is used in studies of monocyte maturation and macrophage differentiation.
Dysfunction of CD11b is associated with systemic lupus erythematosus and ITGAM-related susceptibility to autoimmune disease.
