Target Information

The CD3 subunit complex is crucial for transducing antigen-recognition signals into the cytoplasm of T cells and regulating the surface expression of the TCR complex. T cell activation through the TCR involves the cytoplasmic tails of CD3γ, CD3δ, CD3ε, and CD3ζ subunits. These subunits are structurally related immunoglobulin superfamily members encoded by genes on human chromosome 11.

CD3 components have cytoplasmic tails that associate with signal transduction molecules via a double tyrosine-based motif. CD3 plays roles in TCR-induced growth arrest, survival, and proliferation. It is expressed on 68–82% of normal peripheral blood lymphocytes, 65–85% of thymocytes, and Purkinje cells in the cerebellum, but not on B or NK cells.

Reduced T lymphocyte percentages may be seen in some autoimmune diseases. The genes encoding CD3ε, γ, and δ are located on chromosome 11. Mutations in CD3 genes are associated with CD3 immunodeficiency.
