STAT3는 STAT (Signal Transducers and Activators of Transcription) 단백질 계열에 속하며, 수용체 관련 키나아제에 의해 인산화되어 핵으로 이동한 후 전사인자로 작용합니다. STAT3는 Cytokine 및 성장인자 반응에 의해 활성화되며, CREB-binding protein과 같은 공동활성화 인자가 필요합니다. 또한 Interferon-α, Interferon-γ, EGF, PDGF, IL6 등에 의해 활성화될 수 있습니다.
Tyrosine 705의 인산화는 STAT3 이합체 형성, 핵 이동, DNA 결합 활성에 필수적입니다. 인간에서 STAT3 유전자는 염색체 17의 q arm에 위치하며, 다양한 세포 자극에 대한 유전자 발현을 매개하여 세포 성장과 세포사멸 조절에 중요한 역할을 합니다.
또한 Rac1 GTPase가 STAT3의 활성을 조절하고, PIAS3 단백질은 STAT3의 특이적 억제 인자로 알려져 있습니다. STAT3 유전자의 결손은 배아 초기 단계에서 치명적임이 보고되었습니다.
