Product Specific Information

Description:
AD2 monoclonal antibody reacts with human CD73, a 5′-ectonucleotidase that converts 5′-adenosine monophosphate to adenosine. CD73 is expressed on endothelial cells, B cells, and T cells, including some CD4⁺Foxp3⁺ regulatory T cells. Adenosine production by these cells can inhibit CD4 T cell effector functions such as proliferation and cytokine secretion.

Applications Reported:
This AD2 antibody is reported for use in flow cytometric analysis.

Applications Tested:
Pre-diluted and tested by flow cytometric analysis of normal human peripheral blood cells. Recommended usage is 5 µL (0.125 µg) per test in a final volume of 100 µL. Suitable for 10⁵–10⁸ cells/test.

Fluorochrome Information:
APC-eFluor 780 emits at 780 nm and is excited with the Red laser (633–647 nm). Ensure the instrument can detect this fluorochrome.

Light Sensitivity:
This tandem dye is sensitive to photo-induced oxidation. Protect vials and stained samples from light.

Fixation:
Samples can be stored in:

• IC Fixation Buffer (Product #00-8222-49)
• 1-step Fix/Lyse Solution (Product #00-5333-57)
  for up to 3 days at 4°C in the dark with minimal impact on brightness and FRET efficiency.

Target Information

CD73 (Ecto-5′-nucleotidase, 5′-ribonucleotide phosphohydrolase) catalyzes the conversion of purine 5′-mononucleotides to nucleosides, preferring AMP as a substrate.
It is a dimer of two 70 kDa subunits bound to the plasma membrane via a glycosyl phosphatidyl inositol linkage.
CD73 serves as a marker for lymphocyte differentiation, and its deficiency is associated with various immunodeficiency diseases.
The CD73 gene is localized to chromosome 6q14–q21. Mutations can lead to calcification of joints and arteries and intestinal tuberculosis.
Two transcript variants encoding different isoforms have been identified.
