Raf-1 (c-Raf)은 MAP kinase kinase kinase (MAP3K)로, 막 결합 GTPase인 Ras 패밀리 단백질의 하위에서 작용합니다. 활성화된 Raf-1은 MEK1 및 MEK2를 인산화하여 활성화시키며, 이는 다시 ERK1 및 ERK2를 활성화합니다. 활성화된 ERK는 세포 생리의 다양한 과정에 관여하며, 유전자 발현 조절, 세포주기, 세포사멸, 분화, 이동 등에 중요한 역할을 합니다.
이 유전자의 돌연변이는 Noonan syndrome 5 및 LEOPARD syndrome 2와 관련이 있습니다.
