Target Information
Caspase 8 (CASP8)는 cysteine-aspartic acid protease (caspase) 계열에 속하는 단백질로, 세포자멸사(apoptosis)의 실행 단계에서 중심적인 역할을 합니다. Caspase는 비활성 전구체 형태로 존재하며, 활성화 시 내부의 aspartic 잔기에서 절단되어 대소 단위체로 구성된 효소 복합체를 형성합니다.
Caspase 8은 Fas 및 다양한 세포자멸 자극에 의해 유도되는 programmed cell death 과정에 관여하며, FADD 단백질과 상호작용하는 death effector domain(DED)을 포함하고 있습니다.
또한 Huntington 병 환자의 뇌 조직에서 Caspase 8이 불용성 분획에서 검출되어 신경퇴행성 질환과의 연관성이 제시되었습니다. Caspase 8은 FADD의 DED와 결합하고, downstream caspase(예: Caspase 9)를 활성화하여 세포자멸을 유도합니다. 과발현 시 apoptosis를 유도하며, ICE 계열 억제제에 의해 차단될 수 있습니다. 다양한 스플라이싱 변이체가 보고되어 있습니다.
