Product Specific Information

The monoclonal antibody MH9D1 recognizes interleukin-9 (IL-9), a proinflammatory cytokine involved in immune regulation. IL-9 is secreted by Th9 cells, which can be derived from Th2 cells with TGF-β or directly from naïve CD4+ T cells with TGF-β and IL-4.

IL-9 belongs to the common cytokine receptor γ-chain-dependent family, including IL-2, IL-4, IL-7, IL-15, and IL-21. It affects Th2 lymphocytes, B lymphocytes, mast cells, eosinophils, and epithelial cells, and is implicated in asthma and allergy-related disorders.
Staining with MH9D1 is not blocked by MH9A4, indicating distinct epitope recognition.

Applications Reported: Intracellular staining followed by flow cytometric analysis.
Applications Tested: Pre-titrated and validated on restimulated Th2-polarized human PBMCs using the Intracellular Fixation & Permeabilization Buffer Set (Product #88-8824-00).

Target Information

Interleukin 9 (IL-9) is produced by activated Th2 lymphocytes and regulates multiple cell types involved in asthma-related immune responses, including B/T lymphocytes, mast cells, eosinophils, and epithelial cells.
Mouse IL-9 cDNA encodes a 144 amino acid precursor with an 18 amino acid signal peptide, forming a mature 14 kDa glycosylated protein.
TGF-β plays a key role in Th9 differentiation by reprogramming Th2 cells to secrete IL-9.
