Target Information

Proteolytic degradation is critical to maintaining appropriate levels of short-lived and regulatory proteins involved in cellular metabolism, stress response, antigen presentation, and signaling. The ubiquitin-proteasome pathway deconstructs most cytosolic and nuclear proteins, while others are degraded via the vacuolar pathway involving endosomes, lysosomes, and the endoplasmic reticulum. The 26S proteasome, an ATP-dependent multisubunit complex (~2.5 MDa), consists of a 20S core and 19S regulatory subunits that recognize ubiquitinated proteins, unfold them, and translocate them for degradation. Defects in this pathway are linked to several genetic diseases such as cystic fibrosis, Angelman’s syndrome, and Liddle syndrome.
