Proteolytic degradation is essential for maintaining proper levels of short-lived and regulatory proteins involved in diverse cellular processes such as metabolism, stress response, antigen presentation, receptor modulation, ion channel regulation, cell cycle control, transcription, and signaling.
The ubiquitin-proteasome pathway degrades most cytosolic and nuclear proteins in eukaryotic cells, while others are processed via the vacuolar pathway involving endosomes, lysosomes, and the endoplasmic reticulum.
The 26S proteasome is an ATP-dependent, multisubunit complex (~2.5 MDa) composed of a 20S proteolytic core and one or two 19S regulatory subunits. The 19S units recognize ubiquitinated proteins, unfold them, and translocate them into the 20S core for degradation.
Defects in this pathway are associated with several genetic diseases, including cystic fibrosis, Angelman’s syndrome, and Liddle syndrome.
