Target Information
DNA methylation, or the addition of methyl groups to cytosine bases in the dinucleotide CpG, is essential for proper development and gene expression regulation. The methylation pattern involves enzymatic methylation and demethylation. The mammalian demethylation enzyme exhibits demethylase activity associated with a methyl-CpG-binding domain (MBD).
MeCP2 and MBD1 repress transcription by binding specifically to methylated DNA. MBD2 and MBD4 (also known as MED1) colocalize with heavily methylated satellite DNA and mediate methylation signal functions. MBD3 does not bind methylated DNA. MBD1, MBD2, MBD3, and MBD4 are expressed in somatic tissues, but MBD1 and MBD2 expression is reduced or absent in embryonic stem cells. MBD4 shares homology with bacterial base excision repair DNA N-glycosylases/lyases and is sometimes mutated in microsatellite unstable tumors.
