Target Information
TRF1은 텔로미어 길이 조절의 음성 조절자로 알려져 있습니다. 최근 TRF1에 결합하는 단백질로 tankyrase와 Tin2가 확인되었습니다. Tankyrase는 ankyrin 반복 서열과 poly(ADP-ribose) polymerase (PARP) 촉매 도메인과의 상동성을 가지며, TRF1과 결합하여 텔로미어에 위치합니다. TRF1의 ADP-리보실화는 텔로미어 DNA 결합 능력을 감소시켜, 인간 세포의 텔로미어 기능이 poly(ADP)-리보실화에 의해 조절됨을 시사합니다.
TIN2 단백질의 cDNA는 yeast two-hybrid 라이브러리 스크리닝을 통해 TRF1의 상호작용 파트너로 확인되었습니다. Tin2는 텔로미어에 위치하며, 텔로미어 길이의 적절한 조절에 필수적입니다. TIN2와 TRF1은 모두 널리 발현되어 있으며, 텔로머라아제에 의한 텔로미어 연장을 억제하는 데 함께 작용합니다. 그러나 내인성 TIN2 발현 수준은 낮아, 특정 세포 시스템에서는 면역염색으로 관찰하기 어려울 수 있습니다.
