For reconstitution, add 100 µL distilled water to achieve a final antibody concentration of approximately 1 mg/mL.
For conjugation experiments, perform an additional desalting step using Zeba Spin Desalting Columns (7K MWCO, 0.5 mL, Product #89882).
Target Information
CD33 is a transmembrane protein of the Siglec family, containing immunoreceptor tyrosine-based inhibitory motifs (ITIMs) that recruit SHP-1 and SHP-2 phosphatases. These motifs also mediate ubiquitin-dependent receptor removal from the cell surface.
CD33 is expressed on myelomonocytic cells, binds sialic acid residues in N- and O-glycans, and serves as a therapeutic target for acute myeloid leukemia. It is found on granulocyte and macrophage precursors in bone marrow but not on pluripotent stem cells. CD33 is also expressed on peripheral monocytes and is a useful marker for distinguishing myelogenous leukemia cells from lymphoid or erythroid leukemias.
Diseases associated with CD33 dysfunction include gallbladder lymphoma and extracutaneous mastocytoma.
