Target Information
TNK2 (ACK, activated Cdc42-associated kinase) is a non-receptor tyrosine kinase that signals downstream of Cdc42. TNK2 is involved in tumorigenesis and metastasis, particularly in prostate cancer. It mediates CDC42-dependent cell migration via phosphorylation of BCAR1 and may play roles in synaptic function, plasticity, and brain development. TNK2 activates AKT1 by phosphorylating Tyr-176 and phosphorylates AR on Tyr-267 and Tyr-363, promoting recruitment to androgen-responsive enhancers. It also phosphorylates WWOX (Tyr-287) and MCF2, enhancing its GEF activity toward Rho family proteins. TNK2 contributes to AXL receptor regulation and confers metastatic properties by modulating tumor suppressors and pro-survival factors.
