Target Information
MDM2는 p53의 유비퀴틴 리가아제로, p53 안정성 조절에 중심적인 역할을 합니다. 인간 염색체 12번 q arm에 위치하며, Akt에 의해 Ser166 및 Ser186에서 인산화되면 p300과의 상호작용이 증가하여 MDM2 매개 p53 유비퀴틴화 및 분해를 촉진합니다. 또한 MDM2의 인산화는 p19ARF와의 결합을 차단하여 p53 분해를 증가시킵니다.
MDM2는 p53 기능을 음성 조절하며, 과발현 시 p53 종양 억제 기능이 감소할 수 있습니다. 이 단백질은 E3 유비퀴틴 리가아제 활성을 통해 p53을 분해할 수 있으며, 세포사멸(apoptosis)과 세포 주기 조절에도 관여합니다.
MDM2는 연부 조직암 및 유방암을 포함한 다양한 인간 악성종양에서 과발현되며, p53 외에도 retinoblastoma 1 및 ribosomal protein L5 등과 상호작용하여 세포 주기, 세포사멸, 종양발생 과정에 관여합니다. 40개 이상의 대체 스플라이싱 변이체가 보고되어 있습니다.
