Target Information
TRPC6은 세포막에서 수용체 활성화 칼슘 채널을 형성하는 단백질입니다. 이 채널은 diacylglycerol(DAG)에 의해 활성화되며, phosphatidylinositol 신호전달 시스템의 제어를 받는 것으로 알려져 있습니다. TRPC6의 활성은 protein kinase C에 의존하지 않으며, 낮은 세포 내 칼슘 농도에 의해 유도되지 않습니다.
TRPC6 유전자의 결함은 Focal Segmental Glomerulosclerosis 2 (FSGS2)의 원인으로 알려져 있으며, TRPC6은 포유류 TRP(superfamily) 중 “canonical TRP (TRPC)” 계열에 속합니다. TRPC6/6/7 아형은 DAG 유사체에 의해 활성화될 수 있으며, PLC-coupled receptor에 의해 조절되는 양이온 채널의 구성 요소로 작용합니다.
TRPC6은 DAG에 의해 PKC 비의존적으로 활성화되며, IP3 활성화에는 반응하지 않습니다. 생체 내에서 DAG-활성화 수용체 작동 양이온 채널로 기능한다는 증거가 증가하고 있습니다. TRPC6 관련 질환으로는 Glomerulosclerosis, Focal Segmental, 2 및 Focal Segmental Glomerulosclerosis가 있습니다.
