Description:
The A12 monoclonal antibody reacts with human CD150, an approximately 70 kDa transmembrane glycoprotein also known as Signaling Lymphocyte Activation Molecule (SLAM). CD150 is expressed by T and B cells and is rapidly upregulated upon activation. Immature thymocytes and dendritic cells also express this antigen. Signaling through SLAM induces proliferation and cytokine production of T cells and plays a role in activation of T and B cells.

This A12 (7D4) antibody has been reported for use in flow cytometric analysis.

Applications Tested:
The antibody has been pre-titrated and tested by flow cytometric analysis of normal human peripheral blood cells. Recommended use is 5 µL (1 µg)/test in a final volume of 100 µL. The number of cells can range from 10⁵ to 10⁸ cells/test.

Target Information

CD150 (SLAM, Signaling Lymphocyte Activation Molecule) belongs to the immunoglobulin gene superfamily and is involved in T cell stimulation. SLAM is constitutively expressed on peripheral blood memory T cells, T cell clones, immature thymocytes, and a proportion of B cells, and is rapidly induced on naive T cells after activation. Activated B cells express membrane-bound, soluble, and cytoplasmic isoforms of SLAM.

SLAM-SLAM homophilic interactions during B–B and B–T cell communication enhance proliferation and differentiation of activated B cells. CD150 also serves as a receptor for the measles virus and acts as a co-activator on T and B cells. It interacts with SH2D1A and PTPN11 via its cytoplasmic domain. Mutations in the CD150 gene may be associated with X-linked lymphoproliferative disease (XLP).
