Product Specific Information

The HI100 monoclonal antibody reacts with human CD45RA, a 220 kDa molecule expressed by subpopulations of CD4+ and CD8+ peripheral T lymphocytes, and B cells. CD45RA+ T cell populations are mainly naive/virgin, allowing the HI100 mAb to serve as a phenotypic marker to discriminate T cell subsets.

• Applications Tested: Pre-titrated and tested by flow cytometry using normal human peripheral blood cells
• Recommended Use: 5 µL (0.8 µg) per 100 µL sample (10⁵–10⁸ cells/test)

Each product includes:

• 1 vial of CellBlox Plus Blocking Buffer

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability analysis.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
• Add 5 µL of CellBlox Plus Blocking Buffer per sample (regardless of number of NovaFluors).

NovaFluor conjugates utilize Phiton™ technology, employing nucleic acid dye structures for engineered fluorescent signatures with minimal spillover and spread.

Target Information

CD45RA is an isoform of the CD45 complex with restricted expression among lymphoid subtypes. CD45 (LCA, leukocyte common antigen) is a receptor-type protein tyrosine phosphatase expressed on all nucleated hematopoietic cells, comprising ~10% of lymphocyte surface proteins.
It plays a key role in lymphocyte development, antigen signaling, and regulation of Src-family kinases.
CD45 isoforms differ by alternative splicing, affecting extracellular domains but sharing identical transmembrane and cytoplasmic regions.
CD45 is absent in mature red cells and non-hematopoietic tissues. Dysfunction is associated with severe combined immunodeficiency and Hepatitis C.
