Product Specific Information

The HI100 monoclonal antibody reacts with human CD45RA, a 220 kDa molecule expressed by subpopulations of CD4+ and CD8+ peripheral T lymphocytes, and B cells. CD45RA+ T cell populations are mainly naive, allowing discrimination of T cell subsets.

• Applications Reported: Flow cytometric analysis
• Applications Tested: Pre-titrated and validated for flow cytometry of normal human peripheral blood cells.
• Recommended Usage: 5 µL (0.8 µg) per test in 100 µL final staining volume; cell count range 10⁵–10⁸ cells/test.
• Included Components: 1 vial of NovaFluor conjugate and 1 vial of CellBlox Plus Blocking Buffer.

NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assessment with NovaFluor dyes.

CellBlox Plus Blocking Buffer

This conjugate ships with CellBlox Plus Blocking Buffer (Cat. No. C001T06F01), offering improved formulation for optimal staining.
Add 5 µL per sample to antibody cocktails before combining with cells. For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells).

NovaFluor Technology

NovaFluor conjugates utilize Phiton™ technology, employing nucleic acid dye structures for engineered fluorescent signatures with reduced spillover and spread.
Excitation: 408 nm; Emission: 800 nm; Laser: 488 nm (Blue).

Target Information

CD45RA is an isoform of the CD45 complex, expressed selectively among lymphoid cell subtypes. CD45 (LCA) is a receptor-type protein tyrosine phosphatase found on all nucleated hematopoietic cells, comprising ~10% of lymphocyte surface proteins.
It regulates Src-family kinases and plays key roles in lymphocyte development, antigen signaling, integrin-mediated survival signaling, and apoptosis.
CD45 dysfunction is associated with conditions such as severe combined immunodeficiency and Hepatitis C.
