Product Specific Information

The HI100 monoclonal antibody reacts with human CD45RA, a 220 kDa molecule expressed by subpopulations of CD4+ and CD8+ T lymphocytes and B cells. CD45RA+ T cell populations are mainly naïve/virgin, allowing the use of HI100 mAb as a phenotypic marker to discriminate T cell subsets.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested by flow cytometric analysis of normal human peripheral blood cells. Use at 5 µL (0.8 µg) per test (100 µL final volume). Cell number range: 10⁵–10⁸ cells/test.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability with NovaFluor dyes.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) must be used for optimal staining.
• Add 5 µL of CellBlox Plus Blocking Buffer per sample before combining with cells.
• For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells).

Based on Phiton™ technology utilizing novel nucleic acid dye structures engineered for optimized fluorescent signatures, minimizing spillover and spread.

Target Information

CD45RA is an isoform of the CD45 complex, a receptor-type protein tyrosine phosphatase expressed on all nucleated hematopoietic cells except mature red cells. It regulates Src-family kinases and plays a key role in lymphocyte development, antigen signaling, and integrin-mediated survival pathways. CD45 isoforms differ by extracellular domain composition due to alternative splicing and are expressed variably by cell type and physiological state. CD45 dysfunction is associated with severe combined immunodeficiency and Hepatitis C.
