• For immunohistochemistry on PFA-fixed tissues: incubate antibody for 30 minutes at room temperature.
• For formalin-fixed tissues: heat sections in 10 mM Tris with 1 mM EDTA (pH 9.0) for 45 minutes at 95°C, then cool at room temperature for 20 minutes before staining.

Target Information

Neurofilaments are intermediate filament proteins found specifically in neurons and are composed of three major subunits: NF-L, NF-M, and NF-H.

• NF-L (Light chain): Runs at approximately 70 kDa on SDS-PAGE.
• NF-M (Medium chain): Intermediate molecular weight.
• NF-H (Heavy chain): Runs at 180–220 kDa depending on species.
  These proteins are essential for maintaining neuronal caliber and are developmentally and tissue-specifically regulated.
