Product Specific Information

This NKTA255 monoclonal antibody reacts with human CD305 (LAIR1), a type I transmembrane glycoprotein possessing an extracellular Ig-like domain and two intracellular ITIMs. CD305 is expressed on most immune cells including T, NK, B cells, monocytes, dendritic cells, eosinophils, basophils, and mast cells. It is also found on CD34+ progenitor cells and thymocytes.

As an inhibitory receptor, CD305 blocks cytokine-mediated signaling, cell proliferation, and apoptosis. Collagens have been identified as its ligands. The NKTA255 antibody has been reported to inhibit NK cytolytic activity.

Applications Tested: Pre-titrated and tested on human peripheral blood cells
Use at 5 µL (0.4 µg) per test (100 µL final volume). Cell number may range from 10⁵ to 10⁸ cells/test.

Each product includes:

  • 1 vial of NovaFluor conjugate
  • 1 vial of CellBlox Plus Blocking Buffer

Important Usage Notes:

  • NovaFluor dyes are incompatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
  • Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assays.
  • CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining with NovaFluor conjugates.
  • Add 5 µL per sample to antibody cocktails prior to combining with cells.

NovaFluor conjugates utilize Phiton™ technology with engineered fluorescent signatures for minimal spillover and spectral spread.
Excitation: 637 nm; Emission: 685 nm; Laser: 633–640 nm (Red).


Target Information

LAIR1 (CD305) acts as an inhibitory receptor regulating NK, B, and T cell cytolytic functions. Upon Tyr phosphorylation, it recruits phosphatases PTPN6 and PTPN11, reducing intracellular calcium increase from B-cell receptor signaling. It modulates cytokine production in CD4+ T-cells (down-regulating IL2 and IFNG, inducing TGF-β), and decreases IgG/IgE production in B-cells along with IL8, IL10, and TNF secretion. LAIR1 also inhibits proliferation, induces apoptosis in myeloid leukemia cells, and prevents NF-κB activation. It suppresses differentiation of peripheral precursors into dendritic cells.
