Product Specific Information

This NKTA255 monoclonal antibody reacts with human CD305 (LAIR1), a type I transmembrane glycoprotein with an extracellular Ig-like domain and two intracellular ITIMs. CD305 is expressed on most immune cells, including T, NK, B cells, monocytes, dendritic cells, eosinophils, basophils, and mast cells. It is also found on CD34+ hematopoietic progenitor cells and thymocytes. Several isoforms exist due to alternative splicing, with expression varying by cell type.

As an inhibitory receptor, CD305 blocks cytokine-mediated signaling, cell proliferation, and apoptosis. Collagens serve as ligands for CD305. The NKTA255 antibody has been reported to inhibit NK cytolytic activity.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and validated for human peripheral blood cells.
Use at 5 µL (0.4 µg) per test in a final volume of 100 µL. Cell number can range from 10⁵ to 10⁸ cells/test.

Important Notes:

• NovaFluor dyes are incompatible with DNA intercalating viability dyes such as propidium iodide, 7-AAD, and DAPI.
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assays.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) must be used for optimal staining.
  • Add 5 µL per sample to antibody cocktails before combining with cells.
  • For single-color controls, use 5 µL per 100 µL of cell sample (10³–10⁸ cells).

Target Information

LAIR1 (CD305) functions as an inhibitory receptor that negatively regulates cytolytic activity of NK, B, and T cells. Upon activation by tyrosine phosphorylation, it recruits phosphatases PTPN6 and PTPN11, reducing intracellular calcium increases triggered by B-cell receptor ligation. It modulates cytokine production in CD4+ T-cells, suppressing IL2 and IFNG while inducing TGF-β secretion. LAIR1 also downregulates IgG and IgE production in B-cells and inhibits IL8, IL10, and TNF secretion. In myeloid leukemia cells, it inhibits proliferation, induces apoptosis, and prevents NF-κB activation and dendritic cell differentiation.
