Immunohistochemistry (PFA fixed):

• Incubate antibody for 30 min at room temperature (RT).
• For formalin-fixed tissues, heat tissue sections in 10 mM Tris with 1 mM EDTA (pH 9.0) for 45 min at 95°C, followed by cooling at RT for 20 min.

Target Information

Cytokeratins (CK) are intermediate filaments of epithelial cells found in both keratinizing (e.g., skin) and non-keratinizing tissues (e.g., mesothelial cells). Although not typical endothelial markers, cytokeratins are present in some microvascular endothelial cells.
There are at least 20 cytokeratin types (40–70 kDa, pI 5–8.5), identified by 2D gel electrophoresis.
Cytokeratins are classified into two groups:

• Type I (acidic polypeptides)
• Type II (basic polypeptides)

Every epithelial cell expresses at least one member from each class.
Monoclonal antibodies to cytokeratin proteins are valuable tools for tumor identification and classification.
