Target Information
Tau is a neuronal microtubule-associated protein found predominantly on axons. It promotes tubulin polymerization and stabilizes microtubules. The C-terminus binds axonal microtubules, while the N-terminus associates with neural plasma membrane components, functioning as a linker between the cytoskeleton and membrane structures.
Tau localization defines axonal polarity in neurons. Short isoforms contribute to cytoskeletal plasticity, whereas longer isoforms stabilize microtubules. In its hyper-phosphorylated form, Tau is the major component of paired helical filaments (PHF), which form neurofibrillary tangles characteristic of Alzheimer’s disease (AD). Hyper-phosphorylation disrupts microtubule binding, leading to neuronal degeneration. Multiple kinases, including GSK-3β, PKA, cdk5, and casein kinase II, are known to phosphorylate Tau. Abnormal Tau phosphorylation is implicated in AD and other neurodegenerative diseases such as Pick’s disease, frontotemporal dementia, corticobasal degeneration, and progressive supranuclear palsy.
