Target Information
CD31 (platelet endothelial cell adhesion molecule-1, PECAM-1) is an inhibitory coreceptor regulating T and B cell signaling via dual ITIM motifs. Upon phosphorylation by kinases, these motifs serve as docking sites for protein-tyrosine phosphatases.
CD31 is widely expressed in the vascular compartment, primarily at cell junctions. Its N-terminal Ig-like domain mediates homophilic binding, essential for cell-cell interactions.
CD31 plays diverse roles in:
- Integrin-mediated cell adhesion
- Transendothelial migration
- Angiogenesis and apoptosis
- Negative regulation of immunoreceptor signaling
- Autoimmunity and macrophage phagocytosis
- IgE-mediated anaphylaxis and thrombosis
