Target Information

Calpain-6 (also known as calpamodulin or calpain-X) is an intracellular, calcium-dependent cysteine protease. Unlike the ubiquitous calpain-1 and calpain-2, calpain-6 exhibits tissue-specific expression, primarily in the placenta, though it is expressed embryonically in various tissues.
The classical calpain family consists of a small common subunit (calpain-4) and a large variable subunit. It is unclear whether calpain-6 associates with a small subunit. The large subunit includes domain-I (amino terminal), domain-II (proteinase), domain-III, and domain-IV (EF-hand domain, or domain T in calpains 5 and 6).
Calpain-6, located on the X chromosome, lacks the EF-hand calcium-binding motif found in classical calpains. Additionally, its canonical active site residues (Cys, His, Asn) are replaced by Lys, His, Asn in humans and Lys, Tyr, Asn in mice, suggesting that calpain-6 is likely proteolytically inactive.
