Description:
The 104 monoclonal antibody reacts with the mouse CD45 molecule, the leukocyte common antigen (LCA) in CD45.2-expressing mouse strains. These include C57BL/6, BALB/c, C58, DBA/1, DBA/2, C3H/He, CBA, 129, A, and AKR strains. CD45.2 is expressed by all leukocytes in these strains.
Applications Reported:
The 104 antibody has been reported for use in flow cytometric analysis and immunohistochemical staining of frozen tissue sections.
Applications Tested:
The 104 antibody has been tested by flow cytometric analysis of mouse splenocytes. Recommended usage is ≤0.5 µg per test (100 µL final volume). Cell number should be determined empirically (10^5–10^8 cells/test). Optimal titration is recommended for each assay.

0.2 µm post-manufacturing filtered.
Target Information
CD45.2 (LCA, leukocyte common antigen) is a receptor-type protein tyrosine phosphatase expressed in all nucleated hematopoietic cells, comprising ~10% of surface proteins in lymphocytes. It is crucial for lymphocyte development and antigen signaling through regulation of Src-family kinases.
CD45.2 exists as multiple isoforms due to alternative splicing, differing in extracellular domains but sharing identical transmembrane and cytoplasmic regions. Isoform expression varies by cell type and physiological state.
Beyond immunoreceptor signaling, CD45.2 also promotes cell survival via integrin-mediated pathways and participates in DNA fragmentation during apoptosis. CD45RA is an isoform with restricted expression among lymphoid subsets.
