Target Information
Focal Adhesion Kinase (FAK)는 125 kDa의 비수용체 단백질 티로신 키나아제로, Src의 기질로 작용하며 인테그린 신호전달의 핵심 요소입니다. FAK는 세포 확산, 분화, 이동, 세포사, 세포주기 G1에서 S기로의 전환 등 다양한 세포 기능에 중요한 역할을 합니다.
FAK는 중심 촉매 도메인과 C-말단 꼬리를 가지며, 이는 세포가 인테그린 수용체를 통해 세포외기질에 부착하는 초점부착(focal adhesion) 부위에 위치하도록 합니다.
Fibronectin과의 결합 시 FAK의 티로신 인산화가 증가하며, Grb2 및 c-Src PTK와의 결합을 통해 ERK2 MAP kinase가 활성화됩니다. FAK 유전자는 여러 전사 변이체를 가지며, 그중 세 가지의 full-length 형태가 확인되었습니다.
