• For IHC (PFA fixed): Incubate antibody for 30 minutes at room temperature.
• Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 minutes at 95°C, followed by cooling at room temperature for 20 minutes.

Target Information

Alpha 2-Macroglobulin (A2M) is an approximately 720 kDa homotetrameric glycoprotein composed of four identical ~180 kDa subunits. It shares with other alpha-macroglobulins (such as complement components C3, C4, and pregnancy zone protein PZP) a broad binding capacity for various ligands, functioning as a humoral defense barrier against foreign peptides in plasma. A2M can interact with and capture nearly any proteinase, acting as a panprotease inhibitor. In Alzheimer’s disease (AD) patients, A2M has been localized to diffuse amyloid plaques, suggesting a role in AD etiopathology.
