DMPK는 근육 및 신경 조직에서 발현되는 serine/threonine kinase로, untranslated region의 돌연변이는 근긴장성 이영양증(myotonic dystrophy)의 특징입니다. 이 유전자가 암호화하는 단백질은 Rho 계열의 소형 GTPase와 상호작용하는 다른 kinase와 밀접하게 관련되어 있습니다. 이 효소의 기질에는 myogenin, L-type 칼슘 채널의 β-소단위, phospholemman 등이 포함됩니다.
3' untranslated region에는 CTG 삼핵산 반복서열이 5–37회 존재하며, 이 불안정한 서열이 50–5,000회로 확장되면 근긴장성 이영양증 I형을 유발하며 반복 수가 증가할수록 질환의 심각도가 증가합니다. 반복 확장은 국소 크로마틴 구조의 응축을 유발하여 해당 영역 유전자 발현을 방해합니다.
