The Human IL-10 Simplex ProcartaPlex Kit measures IL-10 protein and is designed to be combinable with other Simplex kits, allowing you to create your own multiplex panel utilizing Luminex xMAP technology for protein detection and quantitation. When combining multiple Simplex kits, only one buffer kit (sold separately) is needed per assay plate regardless of plex size.

About ProcartaPlex Assays for the Luminex Platform

ProcartaPlex immunoassays are based on the principles of a sandwich ELISA, using two highly specific antibodies binding to different epitopes of one protein to quantitate multiple protein targets simultaneously using a Luminex instrument. These assays require as little as 25 μL of plasma or serum, or 50 μL of cell culture supernatant, and provide analyzed results within four hours.

ProcartaPlex assays utilize Luminex xMAP (multianalyte profiling) technology for simultaneous detection and quantitation of up to 80 protein targets in a single sample. The Luminex beads are internally dyed with precise proportions of red and infrared fluorophores to create spectrally unique signatures recognized by Luminex detection systems (e.g., Luminex 200, FLEXMAP 3D, MAGPIX).

Similar to a sandwich ELISA, matched antibody pairs identify the target protein. Each spectrally unique bead is labeled with antibodies specific for a single target, and bound proteins are detected with biotinylated antibodies and streptavidin–R-phycoerythrin (RPE). This bead-based approach enables multiplex analysis in a single well.

Unlike traditional ELISA, the capture antibody in the ProcartaPlex assay is conjugated to a bead and free-floating in solution, not adsorbed to a microplate well. The Luminex instrument uses dual lasers to distinguish bead signatures and quantify RPE fluorescence, proportional to protein concentration. ProcartaPlex multiplex assays can profile more targets with less sample in the same time as a conventional ELISA.
