The Human IFN-alpha Simplex ProcartaPlex Kit measures IFN-alpha protein and is designed to be combinable with other Simplex kits to create a custom multiplex panel utilizing Luminex xMAP technology for protein detection and quantitation. When combining multiple Simplex kits, only one buffer kit (sold separately) is required per assay plate regardless of plex size.

About ProcartaPlex Assays for the Luminex Platform

ProcartaPlex immunoassays are based on sandwich ELISA principles, using two highly specific antibodies binding to different epitopes of one protein to quantitate multiple protein targets simultaneously via a Luminex instrument.
These assays require as little as 25 μL of plasma or serum, or 50 μL of cell culture supernatant, and just four hours to obtain analyzed results.

ProcartaPlex assays utilize Luminex xMAP (multianalyte profiling) technology for simultaneous detection and quantitation of up to 80 protein targets in a single sample. Each bead is internally dyed with unique fluorophore ratios to create spectrally distinct signatures identifiable by Luminex systems (e.g., Luminex 200, FLEXMAP 3D, MAGPIX).

Similar to a sandwich ELISA, matched antibody pairs identify the protein of interest. Each spectrally unique bead is labeled with antibodies specific for a single target protein, and bound proteins are detected using biotinylated antibodies and streptavidin–R-phycoerythrin (RPE). This enables multiplexed analysis of multiple targets in one well.

The key difference from traditional ELISA is that the capture antibody is conjugated to a bead rather than adsorbed to a microplate well, allowing free-floating reagents. Detection is achieved using two lasers—one for bead identification and the other for RPE fluorescence quantitation—providing protein concentration data proportional to fluorescence intensity.

ProcartaPlex multiplex assays can profile more targets using less sample volume and shorter assay time compared to conventional ELISA.
Panels are available for six species: human, mouse, rat, nonhuman primate, porcine, and canine.
