Target Information
TNK2 (ACK), also known as activated Cdc42-associated kinase, is a non-receptor tyrosine kinase acting downstream of Cdc42. TNK2 participates in tumorigenesis and metastasis, notably in prostate cancer. It mediates CDC42-dependent cell migration via phosphorylation of BCAR1 and may contribute to synaptic function and brain development. TNK2 activates AKT1 by phosphorylating Tyr-176 and phosphorylates AR on Tyr-267 and Tyr-363, promoting recruitment to androgen-responsive enhancers (AREs). It also phosphorylates WWOX (Tyr-287) and MCF2, influencing Rho GTPase signaling. TNK2 regulates AXL receptor levels and contributes to tumor progression by modulating tumor suppressors and pro-survival factors.
