Target Information
Anaplastic Lymphoma Receptor Tyrosine Kinase (ALK) belongs to the insulin receptor superfamily and is vital for brain development. Mutations, rearrangements, and amplifications in the ALK gene have been associated with tumors such as anaplastic large cell lymphoma, neuroblastoma, and non-small cell lung cancer. The most common alteration is chromosomal rearrangement resulting in a fusion gene between ALK and nucleophosmin (NPM). The NPM-ALK fusion product is oncogenic. ALK encodes a receptor tyrosine kinase with a transmembrane and extracellular domain, which are absent in the NPM-ALK fusion product. ALK shows high sequence similarity to LTK and plays a crucial role in neuronal development.
