Target Information

Proteolytic degradation is critical to maintaining appropriate levels of short-lived and regulatory proteins involved in cellular metabolism, stress response, antigen presentation, modulation of receptors and ion channels, cell cycle regulation, transcription, and signaling.
The ubiquitin-proteasome pathway deconstructs most proteins in the eukaryotic cytosol and nucleus, while others are degraded via the vacuolar pathway.
The 26S proteasome is an ATP-dependent multisubunit complex (~2.5 MDa) composed of a 20S core and one or two 19S regulatory subunits. The 19S units recognize ubiquitinated proteins and assist in their unfolding and translocation into the 20S lumen.
Defects in this pathway are associated with diseases such as cystic fibrosis, Angelman’s syndrome, and Liddle syndrome.
