Product Specific Information
Heat-mediated antigen retrieval is recommended prior to staining, using a 10 mM citrate buffer (pH 6.0) for 10–20 minutes followed by cooling at room temperature for 20 minutes.
After antigen retrieval, incubate samples with primary antibody for 30 minutes at room temperature.
A suggested positive control is breast carcinoma.
Target Information
Dihydrofolate reductase converts dihydrofolate into tetrahydrofolate and is required for the synthesis of purines, thymidylic acid, and certain amino acids.
The functional dihydrofolate reductase gene is mapped to chromosome 5, and multiple intronless processed pseudogenes or DHFR-like genes have been identified on separate chromosomes.
Deficiency of dihydrofolate reductase has been linked to megaloblastic anemia.
