• Immunohistochemistry (PFA fixed): Incubate antibody for 30 min at room temperature.
• Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris Buffer with 1 mM EDTA (pH 9.0) for 45 min at 95°C, followed by cooling at room temperature for 20 min.

Target Information

CD34 is a highly glycosylated monomeric surface protein (111–115 kDa) expressed on various stem cell populations. It functions as a stem cell marker with reversible expression on human hematopoietic stem cells. CD34 may act as a surface receptor involved in receptor-mediated endocytosis and regulation of adhesion, differentiation, and proliferation of hematopoietic stem cells and progenitors.

CD34 expression reflects specific stages of hematopoietic development and may mediate stem cell attachment to the bone marrow extracellular matrix or stromal cells. It could also serve as a scaffold for lineage-specific glycans, allowing stem cells to bind lectins expressed by stromal or marrow components. The intracellular domain of CD34 is phosphorylated by activated protein kinase C, suggesting a role in signal transduction.

Diseases associated with CD34 dysfunction include dermatofibrosarcoma and neurofibroma.
