Product Specific Information
Immunohistochemistry (PFA fixed): Incubate antibody for 30 minutes at room temperature.
Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris Buffer with 1 mM EDTA (pH 9.0) for 45 minutes at 95°C, followed by cooling at room temperature for 20 minutes.
Target Information
CD34 is a highly glycosylated monomeric surface protein (111–115 kDa) expressed on various stem cell populations. It serves as a stem cell marker and may function as a surface receptor involved in receptor-mediated endocytosis, adhesion, differentiation, and proliferation of hematopoietic stem cells and progenitors.
CD34 expression reflects specific hematopoietic developmental states with altered adhesion and differentiation properties. It may act as an adhesion molecule facilitating stem cell attachment to bone marrow extracellular matrix or stromal cells and as a scaffold for lineage-specific glycans binding to lectins.
The intracellular domain of CD34 is phosphorylated by activated protein kinase C, suggesting a role in signal transduction.
Diseases associated with CD34 dysfunction include dermatofibrosarcoma and neurofibroma.
