Product Specific Information
The antibody has been negatively preadsorbed using a non-phosphopeptide corresponding to the phosphorylation site to remove antibodies reactive with non-phosphorylated IR/IGF1R.
The final product is purified by affinity chromatography using an IR/IGF1R-derived peptide phosphorylated at tyrosine 1158 (tyrosine 1131 for IGF1R).
CHO-T cells transfected with a vector containing the insulin receptor and stimulated with insulin were used as a positive control.
The immunogenic sequence is conserved in mouse and rat for both IR and IGF1R; this antibody is expected to react with human, mouse, and rat, and has been used in western blotting and immunostaining.
Target Information
Insulin and Insulin-like Growth Factor receptors are transmembrane tyrosine kinase receptors critical for development, cell growth, and metabolism.
Binding of insulin or IGF-1 to the extracellular domain of IR or IGFR triggers autophosphorylation and activation of intrinsic tyrosine kinase activity.
Phosphorylation sites unique to each receptor play key roles in signaling differences.
The catalytic loop within the tyrosine kinase domain contains a three-tyrosine motif: Tyr1158, Tyr1162, and Tyr1163 for IR; Tyr1131, Tyr1135, and Tyr1136 for IGF1R.
Autophosphorylation proceeds sequentially, starting at Tyr1162 (or Tyr1135), followed by Tyr1158 (IGF1R Tyr1131) and Tyr1163 (IGF1R Tyr1136), resulting in full activation of IR and IGF1R.
