Target Information

HIF1-alpha (HIF1A) is a subunit of HIF1, a transcription factor found in mammalian cells under reduced oxygen tension. HIF-1 is a heterodimer consisting of alpha and beta subunits, both belonging to the basic-helix-loop-helix Per-aryl hydrocarbon receptor nuclear translocator-Sim (PAS) family.
HIF1 functions as a transcriptional regulator of the adaptive response to hypoxia. Under hypoxic conditions, HIF-1 activates transcription of over 40 genes, including erythropoietin, glucose transporters, glycolytic enzymes, VEGF, and other genes that enhance oxygen delivery or metabolic adaptation.
HIF1-alpha regulates hypoxia-mediated apoptosis, cell proliferation, and tumor angiogenesis. It interacts with p53 and MDM2 pathways affecting protein degradation under hypoxia. Induction of NOX4 by HIF1-alpha contributes to maintaining ROS levels and hypoxia-induced proliferation.
In humans, HIF1A is located on chromosome 14q. The C-terminal of HIF1A binds to p300, forming complexes that induce hypoxia-responsive genes such as VEGF. Hypoxia plays a role in diseases including myocardial and cerebral ischemia, cancer, pulmonary hypertension, congenital heart disease, and COPD.
