Product Specific Information

The SJ25C1 monoclonal antibody reacts with human CD19, a 95 kDa transmembrane glycoprotein expressed by B cells throughout development except terminal plasma cells. It forms a multimolecular complex with CD21, CD81, Leu13, and MHC class II, associated with the BCR. Signaling through CD19 induces tyrosine phosphorylation, calcium flux, and B cell proliferation. SJ25C1 and HIB19 antibodies recognize overlapping epitopes.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and validated for normal human peripheral blood cells at 5 µL (0.8 µg)/test.
A test is defined as staining a cell sample in 100 µL volume; cell count may vary from 10⁵ to 10⁸ cells/test. Optimize antibody titration for best performance.

Compatibility Notes:
NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for NovaFluor dyes.

Blocking Buffer Usage:
CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining. Add 5 µL per sample before combining with cells. For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton™ technology with engineered nucleic acid dye structures designed for optimized fluorescence and minimal spillover.

Target Information

CD19 is a member of the immunoglobulin superfamily with two Ig-like domains, expressed on all peripheral B cells. It is also found on certain myeloid leukemia cells. CD19 acts as a critical regulator of B cell activation and proliferation, forming part of the B cell receptor complex. Mutations lead to hypogammaglobulinemia, while overexpression causes B cell hyperactivity.
