Heat-mediated antigen retrieval is recommended prior to staining, using a 10 mM citrate buffer (pH 6.0) for 10–20 minutes, followed by cooling at room temperature for 20 minutes. After antigen retrieval, incubate samples with primary antibody for 30 minutes at room temperature. A suggested positive control is placenta or colon tissue.

Target Information

CYR61 is a secreted heparin-binding protein encoded by a growth factor-inducible immediate-early gene. It associates with the extracellular matrix and connective tissue and belongs to the CCN protein family, which includes connective tissue growth factor (CTGF) and the mouse CYR61 homolog Fisp12.

As an angiogenic inducer, CYR61 binds to the cell surface receptor integrin αvβ3, stimulating cell adhesion, migration, and DNA synthesis in human vascular endothelial cells. Expression of CYR61 is elevated during vessel growth, wound healing, and chondrocyte differentiation. CYR61 is also detected in various tumors, where it promotes tumor growth and serves as a marker of tumor progression.
