Target Information

Shiga-like toxins (SLTs), also known as Verotoxins, are produced by enterohemorrhagic Escherichia coli (EHEC) strains such as O157:H7, O26:H11, and O111:H8. These strains are associated with hemorrhagic colitis and hemolytic-uremic syndrome (HUS), characterized by acute renal failure, thrombocytopenia, and microangiopathic hemolytic anemia.

There are several distinct E. coli SLTs:

  • SLT-I and SLT-II are produced by EHEC.
  • SLT-I shares 99% deduced amino acid sequence homology with Shiga toxin.
  • SLT-I and SLT-II share approximately 60% sequence homology and are antigenically distinct.
  • Antibodies to SLT-II can also neutralize a variant, SLT-IIv (SLT-Iiv), produced by E. coli strains causing edema.
    SLT-IIv is cytotoxic for Vero cells but not HeLa cells, distinguishing it from SLT-II.

The toxin structure consists of two domains:

  • A polypeptide: inhibits protein synthesis by targeting ribosomes.
  • B polypeptide pentamer: binds to the eukaryotic cell receptor globotriaosylceramide (Gb3), leading to receptor-mediated endocytosis.
