Application Notes:

• Flow Cytometry: For intracellular staining, activated plasmacytoid dendritic cells were fixed with 2% paraformaldehyde and permeabilized with 0.5% saponin. All steps were performed in presence of 0.5 M EDTA to avoid cell aggregation.
• Western Blot: Tissue was homogenized in loading sample buffer and subjected to SDS-PAGE. The band size is approximately 9 kDa.
• IHC-F: Tissue sections were fixed 10 minutes in acetone and pretreated with avidin/biotin blocking kit. Chronically inflamed liver was used as positive control; normal liver and isotype matched antibody as negative control.

Controls:
Positive control: Activated plasmacytoid dendritic cells
Negative control: Normal liver

Target Information

IP-10 (interferon-gamma induced protein 10 kD; in mouse, also known as cytokine responsive gene 2, Crg-2, or mob-1) is a highly inducible primary response gene belonging to the CXC chemokine superfamily. It was first cloned in 1985. Although its biological functions are not fully understood, IP-10 is known to:

• Stimulate monocyte, NK cell, and T-cell migration
• Regulate T-cell and bone marrow progenitor maturation
• Modulate adhesion molecule expression
• Inhibit angiogenesis

Mig and IP-10 share the same receptor, CXCR3.
