The TRI-COLOR® conjugate is a tandem R-phycoerythrin (PE)-Cy5 conjugate that enables simultaneous multicolor labeling and detection of multiple targets using a single excitation source — the 488 nm line of the argon-ion laser.
Target Information
CD45 (LCA, leukocyte common antigen) is a receptor-type protein tyrosine phosphatase (PTP) expressed in all nucleated hematopoietic cells, accounting for about 10% of lymphocyte surface proteins. It is absent in non-hematopoietic cells and tissues. CD45 plays a critical role in lymphocyte development and antigen signaling by regulating Src-family kinases.
Multiple isoforms of CD45 arise from alternative splicing, differing in extracellular domains but sharing identical transmembrane and cytoplasmic domains. CD45RA, one isoform, shows restricted expression among lymphoid subtypes. CD45 isoforms vary in membrane localization and expression depending on cell type and physiological state.
CD45 is essential for T- and B-cell receptor signaling, suppresses JAK kinases to regulate cytokine signaling, and promotes cell survival through modulation of integrin-mediated pathways, DNA fragmentation during apoptosis, and immune response regulation.
