Target Information
Anaplastic Lymphoma Receptor Tyrosine Kinase (ALK) belongs to the insulin receptor superfamily and is vital for brain development. Mutations, rearrangements, and amplifications in the ALK gene have been found in tumors such as anaplastic large cell lymphoma, neuroblastoma, and non-small cell lung cancer. Chromosomal rearrangement commonly creates a fusion gene between ALK and nucleophosmin (NPM). The NPM-ALK fusion product is oncogenic. ALK is a receptor protein-tyrosine kinase with transmembrane and extracellular domains, which are absent in the NPM-ALK product. ALK shows high sequence similarity to LTK and plays an important role in brain development, affecting specific neurons in the nervous system.
