Target Information

Proteolytic degradation is critical for maintaining appropriate levels of short-lived and regulatory proteins involved in diverse cellular processes such as metabolism, stress response, antigen presentation, receptor modulation, ion channel regulation, cell cycle, transcription, and signaling.
The ubiquitin-proteasome pathway deconstructs most cytosolic and nuclear proteins in eukaryotic cells. Other proteins are degraded via the vacuolar pathway (endosomes, lysosomes, ER). The 26S proteasome (~2.5 MDa) comprises a 20S core complex and 19S regulatory subunits that recognize ubiquitinated proteins and facilitate unfolding and translocation.
The PA28/11S REG activator complex (α, β, γ subunits) enhances 20S core activity.
Defects in this pathway are linked to genetic diseases such as cystic fibrosis, Angelman’s syndrome, and Liddle syndrome.
